目的探讨骨髓间充质干细胞(bonemarrow mesenchymal stem cells,BM-MSC)在造血损伤修复中的作用。方法将同系小鼠股骨、骨壳、注入骨髓条的塑料管及经20Gyγ-射线照射的股骨分别植入同一小鼠腹部皮下,观察1、2、3、4w各移植物造血重建情况。观察髓腔内微循环结构与机能、组织学、分化不同阶段的造血细胞数目与机能等变化,以及骨皮质体外间充质干细胞(MSC)培养与扩增规律。结果乏造血干细胞(HSC)的骨皮质异位移植完全可以造血重建,在移植后4w,移植物已形成完整的封闭的具有正常造血组织的股骨,其重建的程序、速度与具有大量HSC的完整股骨移植的重建基本一致。而经射线破坏和注入塑料管内的骨髓条完全失去造血重建功能。结论骨皮质可能是骨髓MSC的源泉,BM-MSC是造血重建的不可缺少的因素。
Objective To discuss the effect of bone marrow mesenchymal stem cells (BM-MSC) on the hematopoiesis-recovering. Methods The femoral bone with bone marrow, plastic tube with bone marrow plugs and the femoral bone irradiated by 20 Gy γ-irradiation obtained from the syngeneic mice were ectopically transplanted into abdominal subcutaneous of the same mouse, and the hematopoietic-reconstruction of all the graft were observed on 1, 2, 3, 4 week (w) after transplantation. The observation indexes included microcirculation structure and function, histology, the varieties of hematopoietic cells number and function and so on in different differentiated stage and the culture and expansion regulation of MSC derived from femoral bone without marrow. Results The femoral bone without hemopoietic stem cell (HSC) could be hematopoietic-reconstruction completely after ectopic transplantation. The graft formed complete and close femoral bone with normal hematopoietic tissue after 4 weeks of transplantation and the procedure and speed of reconstruction seemed to coincide with the abundant HSC femoral bone. The plastic tube with bone marrow plugs and the irradiated femoral bone lost hematopoietic-reconstruction function completely. Conclusions The femoral bone shaft without bone marrow may be the resource of MSC and BM-MSC is necessary factor in hematopoietic-reconstruction.
