目的:探讨硒含量对大鼠心肌表观遗传因子的作用。方法:使用RT -PCR、Western blot分析等方式,研究硒含量对心肌细胞 TLR2、ICAM1、Gadd45α的表观遗传调控及基因表8达的调节机制。结果:随着硒含量增加,add45α表达、TLR2的表达水平逐渐降低,TLR2启动子甲基化水平将逐渐升高。结论:硒处理能够抑制DNA 异常去甲基化和 TLR2、ICAM1的表达,从而保护大鼠心肌避免炎症损伤。
Objective:To study the selenium keshan disease epigenetic may impact mechanism .Methods :Using RT-PCR and Western blot analysis ,study of selenium content on the myocardial cell TLR2 ,ICAM1 ,epige-netic regulation of Gadd45 alpha and gene expression regulation mechanism .Results :With the selenium content in-creased ,the expression level of add45α,TLR2 expression decreased ,TLR2 promoter Zi Jiaji level will gradually in-crease .Conclusion:The expression of selenium can inhibit the abnormal DNAdemethylation and TLR2 ,ICAM1 , thereby protecting rat myocardial avoid inflammatory injury .
