目的 探究幽门螺杆菌(HP)感染对小鼠十二指肠上皮细胞碳酸氢盐转运蛋白表达的影响。方法 C57小鼠随机分为四组,三组分别给予HP菌株SS1、NCTC11637、NCTC11639感染,一组仅给予同等量的磷酸盐缓冲液(PBS)灌胃(对照组),2 w后收集组织。体外培养十二指肠上皮细胞,与HP菌株共培养,其中对照组给予等量PBS干预,收集细胞。应用实时荧光定量聚合酶链反应(RT-PCR)及Western印迹检测组织及细胞中碳酸氢盐转运蛋白表达的影响。结果 HP感染小鼠后,由吉姆萨染色镜检见,感染后可见大量紫蓝色小体,呈逗点状、短棒状;细胞实验:HP菌株NCTC11637与十二指肠上皮细胞共培养后应用RT-PCR扩增HP16srRNA,小鼠及细胞实验均目的基因为单一扩增,扩增良好。小鼠及细胞实验均3种HP菌种囊性纤维跨膜传导调节因子(CFTR)、离子交换体(SLC)26A3及SLC26A6感染2 w后,碳酸氢盐转运蛋白mRNA及蛋白表达显著低于对照组(P〈0.05)。结论 从体内到体外,HP可通过调控碳酸氢盐转运蛋白含量来干预疾病的进展。
Objective To investigate the influence of bicarbonate transporter proteins expression on duodenal mucosa epithelial cells of the H. pylori (HP) infected mice. Methods C57 mice were randomly divided into four groups. Three groups were given HP strain-SS1, NCTCl1637 and NCTCl1639 to be infected, and the control group was given the same dose of PBS gavage. The organs were collected after 2 weeks. In vitro, duodenal epithelial cells were cultured and given HP strain to be infected, and the control group was given the same dose of PBS. Then, cells were collected. The expression of bicarbonate transporter proteins of organs and cells were detected by RT-PCR and Western blot. Results In animal experiment, after the infection, a large number of purple blue bodies were found, which were funny and short by Giemsa staining. After HP strain-NCTCl1637 infection on duodenal epithelial cells, the amplification of liP 16srRNA was both simple and well by RT-PCR in mice and cells. After 2 weeks of HP strain-SSl, NCTC11637 and NCTC11639 infection on mice, the mRNA and protein expressions of bicarbonate transporter proteins were significantly lower than those of control group in mice and cells. Conclusions In vivo and in vitro, HP can intervene disease progression by regulating the expression of bicarbonate transporter proteins.
