目的本实验利用基因芯片技术检测人工寒潮对于肾血管性高血压大鼠(RHR)外周血白细胞的基因表达谱变化的影响。方法复制RHR经历ACE处理前后尾静脉取血提取白细胞总RNA,经过荧光标记后与5705条基因的oligoDNA芯片杂交、扫描,ACE处理前后样本对照,数据分析筛选出差异表达的基因。结果本实验RHR外周血白细胞基因在5705条基因中共表达4165条。达到显著差异标准表达(ratio≤0.5或者/〉2)的基因共61条(P〈0.01),其中上调18,下调43个。其中转运、转录调控,细胞运动和凋亡相关基因组下调;细胞免疫相关的基因有所上调;运输,信号,代谢。发育和分化组有双向改变。结论利用eDNA芯片技术我们初步发现人工寒潮对于RHR外周血白细胞基因表达有一定影响,可能通过抑制蛋白降解、跨膜转运和免疫反应而减弱应激和炎症反应的能力使机体易于感染,以及通过抑制RHR大鼠血管内膜新生和凝血功能而与卒中前状态相关。
Objective DNA microarrays analysis was used to survey thousands of genes for differential expression trig- gered by artificial cold exposure (ACE) on peripheral white blood cells (WBC) in renovascular hypertensive rats (RHR). Methods Total RNA was extracted from the WBC before and after ACE for mieroarrays analysis using Oiigo-nucleotide arrays (5705 functional genes). Results In total, 61 genes were expressed differentially: 18 genes were up-regulated, 43 genes were down-regulated. The affected genes were distributed among 8 of functional categories (total 12 groups). Transport, Transcription regulator,cell motility,apoptosis related genes are all down-regulated. The cellular immunity related gene Cd3d is up-regulated. Conclusion The ACE effects on WBC gene expression, inhibits the protein degradation, transfer and immunity. The influence on endothelium hyperplasia and coagulation may contribute to the ACE induced stroke.
