为研究丹参提取物对体外培养肝细胞损伤的保护作用,采用胰酶消化法分离原代建鲤肝细胞进行培养,用四氯化碳(CCl4)构建建鲤肝细胞损伤模型,以不同浓度的丹参提取物进行干预,检测损伤肝细胞培养液中丙氨酸转氨酶(GPT)、天冬氨酸转氨酶(GOT)、乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)的活性及丙二醛(MDA)的含量变化。结果表明,以丹参提取物前处理(IV组)中浓度为800μg/mL的丹参提取物效果最好,优于其他两个处理组,与II组相比,显著降低了肝细胞培养液中GOT、GPT、LDH的活性和MDA的含量(P【0.05);显著提高了SOD活性(P【0.05)。这说明丹参提取物对于由CCl4造成的肝细胞损伤具有一定的预防保护作用。
In order to research the protective effect of extracts of Salvia miltiorrhiza on injury of in vitro cultured hepatocytes,the primary cultured hepatocytes of Cyprinus carpiovar Jian were isolated by trypsin digestion method,then the hepatocytes injury model of Cyprinus carpiovar Jian was constructed by carbon tetrachloride(CCl4),and then the hepatocytes were treated with different concentrations of extracts of Salvia miltiorrhiza,in the end,the changes in contents of alanine aminotransferase(GPT),aspartate aminotransferase(GOT),lactate dehydrogenase(LDH),superoxide dismutase(SOD),malondialdehyde(MDA) in culture solution of injury hepatocytes were detected.The results showed that in comparison with the group II,group IV(extracts of Salvia miltiorrhiza at the concentration of 800 μg/mL) could significantly reduce the contents of GOT,GPT,MDA and LDH(P<0.05),but significantly increase the activity of SOD(P< 0.05).Therefore,extracts of Salvia miltiorrhiza had some protective effects on hepatocytes injury induced by CCI4.
