目的研究腺苷(AD)对下丘脑视前正中核(MnPN)神经元调控大鼠睡眠-觉醒周期的影响。方法将SD大鼠随机分为3组:人工脑脊液(ACSF)组、AD组和硝基苯硫嘌呤核苷(NBTI)组,每组5只。采用脑立体定位、微透析和多导睡眠描计技术观察MnPN区给予AD、NBTI对大鼠睡眠-觉醒周期的影响。结果 MnPN区微透析AD后,大鼠非快速眼动(NREM)睡眠增加19.4%(P〈0.01)、觉醒(W)减少26.1%(P〈0.01)、快速眼动(REM)睡眠无明显改变,总睡眠时间(TST)增加17.7%(P〈0.01)。MnPN区微透析NBTI后,大鼠NREM增加11.6%(P〈0.05),W减少16.7%(P〈0.05),REM睡眠无明显改变,TST增加11.0%(P〈0.05)。结论 AD在MnPN区参与睡眠-觉醒周期的调节并具有促睡眠作用。
Objective To investigate the effect of adenosine(AD)in median preoptic mucleus(MnPN)on sleep-wakefulness cycle of rat.Methods Rats were randomly divided into three groups,control group(artificial cerebrospinal fluid group,ACSF),AD group,nitrobenzylthioinosine(NBTI)group.The effects of AD and NBTI,a sensitive AD transporter,on sleep-wakefulness cycle were studied with the methods of brain stereotaxic,microdialysis,polysomngraphy.Results After microdialyzed AD into MnPN of rat,compared with the control group,the ratio of non-rapid eye movement(NREM) sleep was enhanced by 19.4%(P〈0.01),wake(W) was decreased by 26.1%(P〈0.01),total sleep time(TST) was enhanced by 17.7%(P〈0.01),and REM sleep was shown no significant change.After microdialyzed NBTI into MnPN of rat,compared with the control group,the ratio of NREM sleep was enhanced by 11.6%(P〈0.05),W was decreased by 16.7%(P〈0.05),TST was increased by 11.0%(P〈0.05),and it had no effect on REM sleep.Conclusion AD in MnPN takes part in sleep-wakefulness cycle regulation and facilitates sleep.
