目的考察知母有效组分(总皂苷:多糖=1:1)的安全性及其对2型糖尿病大鼠血糖血脂的影响。方法采用急性毒性实验及一般药理学实验,考察知母有效组分的安全性,并通过干预2型糖尿病大鼠(T2DM)模型,考察知母有效组分对其血糖血脂的影响。结果急性毒性实验小鼠无明显染毒反应,药物组小鼠体质量增加,饮食量正常,排泄物未见异常,反应灵活,未见显著的器官病变,知母有效组分的灌胃给药最大耐受剂量为35g·kg^-1,对心血管及呼吸系统无显著作用,知母有效组分高剂量组能显著降低T2DM大鼠模型血糖,改善胰岛素抵抗及血脂紊乱。结论知母有效组分安全性好,具有显著的降低2型糖尿病大鼠的空腹血糖(FBG),糖耐量水平及胰岛素水平(FINS).改善胰岛素抵抗及其血脂紊乱的作用。
Objectives To study the safety of total effective components isolated from Rhizoma Anemarrhenae (Total saponins: Total polysaccharides=l : 1) and its pharmacodynamies of antidiabetes. Methods The safety of total effective components of Rhizoma Anemarrhenae was investigated by acute toxicity test and general pharmacological experiment. The effects of total effective components on the antidiabetic and hyperlipidemia were investigated by interfering with the experimental T2DM rats. Results The the maximum oral administration tolerated dose of total effective components of Rhizoma Anemarrhenae was 35 g ·kg^-1, and no significant influence on central nervous, circulatory and respiratory system was observed. High dose of the total effective components of the Rhizoma Anemarrhenae can significantly reduce blood glucose in T2DM rats, improve insulin resistance and dyslipidemia. Conclusion Total effective components of Rhizoma Anemarrhenae was shown safe and effective on decreasing fasting blood glucose, the glucose tolerance, and insulin levels, improving insulin resistance and dyslipidemia in T2DM rats.
