目的观察扇贝多肽(PCF)对紫外线A(UVA)照射后HaCaT角质形成细胞Fas、c-fos表达的影响,以探讨PCF保护UVA致HaCaT细胞损伤的分子机制。方法体外培养的HaCaT角质形成细胞分为5组:对照组、UVA模型组、5.69mmol/LPCF组、2.84mmol/LPCF组、1.42mmol/LPCF组。蛋白质印迹法检测Fas、c-fos蛋白表达;RT-PCR检测FasmRNA表达;荧光定量PCR检测c-fosmRNA表达。结果 UVA照射后模型组Fas、c-fos表达均显著增加(P〈0.01),1.42~5.69mmol/L剂量范围内的PCF可抑制UVA引起的HaCaT细胞Fas、c-fosmRNA及蛋白表达(P〈0.05),且有剂量依赖性。结论 PCF可通过抑制Fas、c-fos表达而抑制UVA诱导的HaCaT细胞损伤。
Objective To observe the effect of Polypeptide from Chlamys farreri (PCF) on expression levels of Fas and c-fos in HaCaT cells after ultraviolet A (UVA) radiation,and investigate the molecular mechanism of PCF protecting against UVA-induced damage in HaCaT cells.Methods HaCaT cells were divided into five groups: control group,UVA model group,5.69 mmol /L PCF group,2.84 mmol /L PCF group,and 1.42 mmol /L PCF group.Expression levels of Fas and c-fos mRNA was assayed by PCR.Western blot analysis was used to determine the protein levels of Fas and c-fos.Results After UVA exposure,Fas and c-fos expression increased significantly (P〈0.01).1.42 - 5.69 mmol/L PCF inhibited UVA-induced mRNA and protein expressions of Fas and c-fos in HaCaT cells in dose-dependent manner(P〈0.05).Conclusion PCF could attenuate UVA-induced Fas and c-fos expression,which may attribute to its protective effect against UVA-induced damage in HaCaT cells.
