目的探讨Slit—Robo信号对人舌鳞状上皮癌细胞系Tb细胞凋亡的影响及其作用机制。方法以Robol受体胞外区单克隆抗体R5作用于Tb细胞,采用克隆形成实验,流式细胞仪,DNA梯状电泳和Hochest—PI活细胞吸收分析法检测Tb细胞凋亡的发生和凋亡率,并通过Western blot免疫印迹法检测凋亡相关蛋白fas和fasL的表达。结果在R5单克隆抗体作用下,Tb细胞增殖能力减弱,凋亡率增高(61.3%);fas和fasL蛋白表达水平上调,吸光度值分别为2022±243.989和2577±261.222。结论Slit—Robo信号可能抑制口腔癌Tb细胞的凋亡,这种抑制作用可能是通过fas-fasL蛋白的表达调控而实现的。
Objective To study the effect of Slit-Robo signal on apoptosis of human oral squamous cell carcinoma line Tb. Methods After the treatment in Tb cells with monoclonal antibodies (mAb) R5 of against Robol receptor extracellular domain, the apoptosis of Tb cell was examined by clone formation assay, flow cytometry, DNA ladder and Hochest-PI. The expression of fas and fasL proteins was observed by Western blotting analysis. Results After the treatment by R5 mAb, the proliferative rate decreased and the apoptotic rates increased, and the expression of fas and fasL proteins was up-regulated. Condusions SlitRobo signal could inhibit the apoptosis of tumor cells during genesis of tongue cancer by regulating the exDression of fas-fasL Droteins.
