目的研究流行性乙型脑炎DNA疫苗接种局部树突状细胞的形态与功能。方法将含流行性乙型脑炎病毒prM、E蛋白编码基因的DNA疫苗(pJME)肌注免疫BALB/c鼠,HE染色观察接种局部肌肉组织内浸润细胞形态学分布,免疫组化检测CD11b^+、CD11c%+树突状细胞浸润特点,流式细胞仪检测浸润的树突状细胞中CD80^+、CD86^+双阳性细胞的百分比。结果pJME肌注组浸润细胞簇分级(3.0+)、浸润细胞中CD11b^+、CD11c^+树突状细胞百分比分级(2.0+、2.0+)均显著高于空载体肌注组和灭活疫苗肌注组(P〈0.05)。CD80^+、CD86^+双阳性细胞的百分比pJME肌注组为(0.16±0.20)%,空载体组为(0.12±0.30)%,灭活疫苗肌注组为(0.15±0.25)%,树突状细胞CD80、CD86抗原表达无显著差异(P〉0.05)。结论pJME可诱导接种局部多种免疫活性细胞参与,其中有髓样树突状细胞,呈非成熟状态。
Objective To study morphology and function of dendritic cells in injection site induced by DNA vaccine derived from Japanese encephalitis virus. Methods BALB/c mice were vaccinated with DNA vaccine(pJME ) of Japanese Encephalitis virus prM, E encoding protein via inlxamuscular injection. The morphology of local infiltration cells in injection sites was observed through HE staining, and infiltration cells were further studied with the dendritic cell marker CD1 1b and CD1 1c by inmmnohistoehemical methods and with DC mature markers CD80 and CD86 by flow cytometry technique. Results The grading of muscle infiltrates(3.0+) and the grading of CD11b^+ and CD11c^+ cell percentage (2.0+, 2.0+, respectively) produced by pJME via intrmnuscular injection were significantly higher than those by inactivated vaccine and vector (P 〈 0.05), and there was no significant difference in the expressions of CD80 and CD86 on dendritic cells in injection site between pJME (0.16±0.20)%,inactivated vaccine (0.15±0.25)% and vector (0.12±0.30)%(P 〉 0.05). Conclusion pJME can induce many kinds of cell infiltration in injection sites, including immature myeloid dendritic cells.
