猴局部脑缺血模型的构建

目的:研究猕猴脑栓塞模型制作成功后导致颅内压升高,探讨应用七叶皂苷钠处理后猴生命体征的反应及脑脊液压力的改变。方法:本实验将猕猴12只随机分为实验组和对照组,在猕猴脑栓塞模型制作时,实验组给予七叶皂苷钠(0.4mg/kg)20mL,对照组给予0.9%氯化钠20mL。分别记录在栓塞前,栓塞后1、10、30和60min记录血压、心率和呼吸频率的变化;记录脑脊液压力变化。结果:实验组动脉栓塞后1min.收缩压和舒张压较对照组低(P〈0.05):实验组栓塞前后比较血压无明显变化。对照组动脉栓塞后1min,收缩压、舒张压和心率较栓塞前升高(P〈0.05),呼吸频率较栓塞前下降显著(P〈0.05);栓塞后60min.心率较栓塞前下降(P〈0.05)。实验组脑脊液压力在栓塞后与栓塞前比较无差异(P〉0.05),对照组脑脊液压力(CSFP)在栓塞后与栓塞前比较差异有显著性(P〈0.05)。结论:七叶皂苷钠能减少脑动脉栓塞引起急性颅内压增高。降低脑脊液压力,防治脑水肿。

英文摘要:

Objective Observe with sodium aescinate in the preparation of cerebral embolism model process to the influence of the vital signs, and then discusses with sodium aescinate prevention for increased intracranial pressure curative effect. Methods Twelve male macaques randomly were divided into the experimental group and control group, and in the production process of cerebral embolism model, given sodium aescinate (0.4 mg/kg) 20ml, in the control group, given 0.9% sodium cholride 20 mL. Before embolization and 1 min, 10 min, 30 rain,60 min after embolization respectively, the blood pressure, heart rate and breathing rate of change, cerebrospinal fluid pressure values were recorded. Results Group 1 min after embolization, macaques artery systolic blood pressure (SBP) and artery diastolic blood pressure (DBP) than low control group (P 〈 0.05); Compared with the other time before embolization no change happened. In the control group, after embolization 1 min, artery systolic blood pressure and heart rate increased before embolization (P 〈 0.05), artery embolization diastolic blood pressure before increased significantly (P 〈 0.05), breathing down before embolization significant (P 〈 0.05); After embolization 30 min, breathing down before embolization significant (P 〈 0.05); After embolization 60 rain,heart rate is down before embolization (P 〈 0.05). Measurement of cerebrospinal fluid pressure visible. Conclusions With sodium aescinate in the process of cerebral embolism can maintain the smooth vital signs, and improve the intracranial pressure change some of the typical clinical manifestations.
