目的动态研究多巴胺对多巴胺能神经细胞的细胞毒作用。方法用不同浓度的多巴胺处理SH-SY5Y细胞,在一定时程内的不同时间点观察细胞活性,各种氧化应激指标。结果研究发现多巴胺作用于多巴胺能细胞系SH-SY5Y,可导致细胞存活率明显减少,细胞存活率的下降呈时间与浓度依赖;多巴胺处理后,细胞的氧化应激指标OH-自由基、超氧化物岐化酶(SOD)、线粒体膜电位(ΔΨM)、活性氧(ROS)均有不同程度的改变。结论本次实验结果提示多巴胺对多巴胺能细胞的毒性是通过氧化应激途径起作用的。
Objective To study the cytotoxicity of dopamine to dopaminergic SH-SY5Y neuroblastoma cells in a dynamic style. Methods The SH-SY5Y cells were treated with different concentrations of dopamine, and cell viability, as well as the marker of oxidative stress, was observed in a time course. Results Our data showed that dopamine reduced the viability of dopaminergic cell line SHSYSY when dopamine acts on dopaminergic SH-SY5Y cell line. Cellular viability was significantly decreased in a concentration and time dependent manner. After dopamine treatment, the OH-, as well as the superoxide dismutase (SOD), the mitochondrial membrane potential (ΔΨM), and reactive oxidant species (ROS), was significantly changed. Conclusions Our results suggest that the oxidative stress plays a crucial role in the toxic effect of dopamine on dopaminergic cell.
