【目的】建立-种可靠的体外筛选模型评价药物的免疫抑制活性。【方法】在以二硝基氟苯(2,4-dinitrofluorobenzene,DNFB)诱导小鼠耳廓接触性超敏反应(contacthypersensitivity,CHS)模型的基础上,体外培养模型小鼠脾脏T细胞,首先分析细胞增殖活性,进而采用RT—PCR分析核因子IFN-γ、IL-4、T—bet、GATA-3的mRNA表达,最后通过三种临床上常用的免疫抑制药物进行验证。【结果】模型小鼠T淋巴细胞在体外能维持-定时程的高反应性,IFN-γ/IL-4与T—bet/GATA-3比值均明显增加,T细胞向Th1方向分化偏移,同时,环孢素、地塞米松、霉酚酸酯均能显著抑制T细胞的体外增殖。【结论】本研究初步建立了-种简单的可用于筛选免疫制剂的体外细胞模型。
[ Objective] To make a simple and reliable screening model in vitro to investigate the effect of immunosuppressive agents. [ Methods ] DNFB induced CHS mouse model was used. T lymphocytes of the CHS mouse were prepared. Then, proliferation analyse and semi-quantitative RT-PCR were used to explore the characteristics of T lymphocytes from CHS mice. [Results] The T lymphocytes of CHS mice could maintain the higher reactivity in vitro for a period of time, accompanied with the increase of IFN-γ/IL- 4 and T-bet/GATA-3. Furthermore, three kinds of immunosuppressive drugs were proved to inhibit the proliferation of T lymphocytes from CHS mice in vitro. [Conclusion] It's suggested that an easy and feasible screening model for immnnosuppressive agents in vitro might he established preliminarily.
