利用转玉米PEPC基因水稻HFFER(高感水稻白叶枯病)和抗白叶枯病水稻HX-3(具有抗白叶枯病基因Xα25)杂交,获得F1杂种,并对F1杂种进行花药培养,获得42株二倍体植株。通过对这些植株后代进行抗白叶枯病性鉴定,PEPC酶活性和光合速率测定及PCR检测,最后获得同时具有玉米PEPC基因和抗白叶枯病基因Xα25的水稻聚合株系7个,这些株系的农艺性状较HPTER有较大改善,可作为培育高产、抗白叶枯病水稻新品种的种质资源。
A transgenic rice cultivar HPTER, which expresses a maize phosphoenolpyruvate carboxylase (PEPC) gene and is highly susceptible to bacteria blight disease( BB), was crossed with a BB resistant cuhivar HX-3 containing a resistance gene Xα25. By anther culture from the F1 hybrids of the cross, a total of 42 doubled-haploid (DH) regenerated plants were obtained. BB resistance evaluation,PCR analysis and detection of PEPC activity and photosynthetic rate in the DH line led to the identification of 7 pyramid DH lines containing both PEPC gene and Xα25 gene. The pyramid DH lines, with their agronomic traits greatly improved, may act as valuable germplasms for the development of new disease-resistant high-yielding rice varieties.
