目的:鉴定FHL2与Id家族蛋白成员之间的相互作用及分子表位。方法:GST-pulldown方法检测。结果:FHL2与Id家族的4个成员蛋白均存在直接的相互作用关系,FHL2蛋白中的第二个LIM结构域在FHL2/Id相互作用中是必需的,Id蛋白N端结构区域在介导FHL2/Id相互作用中是必需的。结论:FHL2是一个新识别的Id蛋白广谱相互作用因子,FHL2可能参与Id介导的多种生物学效应以及肿瘤发生与进展。
Objective: To investigate the interaction between FHL2 and Id proteins and its corresponding epitope. Methods: Interaction between FHL2 and Id proteins and the corresponding epitope conferring this association were detected by GST pulldown assays. Results: FHL2 directly interacted with all Id proteins. The second LIM domain containing FHI2 or N ter- minal region in Id proteins was essential for the direct binding of FHL2 to ld proteins. Conclusion: FHL2 is a novel and broad interacting factor of the ld proteins. Through cooperating with Id proteins, FHL2 may be involved in multiple biological effects and tumorigenesis mediated by Id proteins.
