目的 探讨过量氟对体外培养山羊成骨细胞生长发育及形态结构的影响。方法 以酶半消化组织块培养法体外分离培养山羊成骨细胞,通过扫描电镜和透射电镜观察不同水平氟对成骨细胞超微结构的影响。结果 扫描电镜观察,1.0×10^-4、5.0×10^-4、1.0×10^-3mol/L氟使成骨细胞突起收缩,胞间连接断裂,胞膜不完整,微绒毛减少。透射电镜观察,1.0×10^-4、5.0×10^-4、1.0×10^-3mol/L氟导致胞膜不完整,胞质浓缩,胞核固缩:细胞染色质固缩、边集;线粒体明显肿胀,嵴断裂、消失或空泡变性;内质网扩张,脱颗粒。结论 过量氟对体外培养的山羊成骨细胞生长发育和形态结构有明显的影响。
Objective In order to investigate the effects of excessive fluoride on the development and morphosis of goat osteoblast in vitro. Methods In the present study, the osteoblasts of caprine fetus, which were obtained from incomplete digestion of long bone periosteum, were cultured in DMEM at 37 ℃. The effects of different doses of fluoride on cellular morphology were investigated using scanning electron microscope (SEM) and transmission electron microscope(TEM). Results Incomplete membrane and shrinkage, detachment or break-up of cell protrusion was observed in the osteoblasts that were cultured with fluoride under 1.0 × 10^-4, 5.0 × 10^-4 and 1.0 × 10^-3 mol/L under the scanning electron microscope. Such uhrastructural features of osteoblasts cultured with fluoride was observed, as cellular membrane disintegration, cytoplasm condensation, nucleus shrinkage, chromatin condensation or peripheral location, mitochondrial obvious swelling, cristal break, vanishment, or vacuolar degeneration, endoplasmic reticulum(ER) expansion or degranulation was observed. Conclusions Excess fluoride has adverse effects on morphology in cultured caprine osteoblasts in vitro.
