目的建立人前脂肪细胞培养及其在药物诱导下向脂肪细胞分化的细胞模型。方法取中国人腹部皮下脂肪组织,经胶原酶消化后在含胎牛血清的DMEM-F12培养基中培养出梭形细胞,以胰岛素、地塞米松及吡咯列酮诱导该梭形细胞向脂肪细胞分化。结果经油红O脂肪染色可见梭形细胞内脂滴的积累。人前脂肪细胞经诱导并继续培养后在旋涡状生长汇合的细胞中心形成一个高密度的细胞岛,该细胞岛由部分可以贴壁的细胞和悬浮的已分化的脂肪细胞组成。结论成熟脂肪组织中存在着具有能分化为脂肪细胞的前脂肪细胞,经适当诱导可定向分化为脂肪细胞。
[Objective] To construct a cell culture model for human preadipocyte differentiation induced by drugs. [Methods] Digested by collagenase Ⅱ, Chinese abdomen subcutaneous adipose tissues were cultured in DMEM - F12 containing FBS to form fusiform cells. Exposed of the ceils to pioglitazone(PIO) along with dexamethasone (DEX) and insulin to induces cell differentiation into adipocytes. [ Results ] Stained by Oil Red O, differentiated adipocytes had accumulation of lipid droplets. There were cell clumps formed in the center of confluence with whirlpool growth after the confluent preadipocytes were induced 2d and then cultured 4d. The cell clumps were so called islets which consist of some attachment cells and some suspending differentiated adipocytes. [ Conclusion] In matured adipocyte tissue there are preadipocytes with potential ability of orientation differentiation into adipocytes by suitable induction.
