目的探讨小柄马勃发酵物对香烟烟雾引起的小鼠慢性阻塞性肺疾病(COPD)的治疗作用及其作用机制。方法采用香烟的烟雾被动熏吸给药法建立慢性阻塞性肺疾病模型。检测细胞因子肿瘤坏死因子-α(TNF-α)白介素-1β和白介素-6(IL-6)的浓度水平,探究小柄马勃发酵物的抗炎作用;测定丙二醛(MDA)和超氧化物歧化酶(SOD)含量评价其抗氧化作用;通过病理切片观察肺的病理状态,考察药物对肺损伤的改善作用;应用蛋白印迹检测慢性阻塞性肺疾病小鼠核因子NF-E2相关因子(Nrf-2)和血红素氧合酶-1(HO-1)的蛋白表达水平。结果小柄马勃的液体发酵物(0.5 g·kg^-1和1 g·kg^-1),显著降低实验小鼠血清中的丙二醛含量,并显著提高超氧化物歧化酶活力、减轻肺泡壁充血和炎性细胞浸润等现象,阻碍核因子NF-E2相关因子和血红素氧合酶-1的表达。结论小柄马勃发酵物(0.5 g·kg^-1和1 g·kg^-1)可能通过其抗氧化作用,改善体内氧化应激条件从而缓解慢性阻塞性肺疾病的疾病进程。
Objective To investigate the effects and mechanism of Lycoperdon pedicellatus Peck.on chronic obstructive pulmonary disease( COPD) induced by cigarette smoke in mice.Methods COPD model was established by passive smoking and smog inhalation of cigarette.the anti-inflammatory effect of Lycoperdon pedicellatus Peck.fermentation was explored by detecting the concentrations of cytokines of TNF-α,IL-1β and IL-6.The antioxidant role was evaluated by determining the content of MDA and SOD.The pathological status of lung was observed by pathological section,and the effect of drugs on lung injury was inspected.Using western blot to detect the protein of Nrf-2 and HO-1 in mice.Results The liquid fermentation of Lycoperdon pedicellatus Peck.( 0.5 g·kg^-1 1 g·kg^-1) could significantly decrease the content of MDA in serum in mice,increase the activity of SOD and could reduce the hyperemia of alveolar wall and inflammatory cell infiltration,with expression of Nrf-2 and HO-1 blocking.Conclusion Lycoperdon pedicellatus Peck.liquid fermentation( 0.5 g·kg^-1 1 g·kg^-1) may improve the oxidative stress in vivo by its antioxidant effect,to alleviating the disease progression of COPD.
