复元醒脑汤对糖尿病脑梗塞大鼠胰岛素抵抗干预作用的实验研究

目的 观察复元醒脑汤对糖尿病脑梗塞大鼠胰岛素抵抗的干预作用。方法 选用SPF 级 SD 雄性大鼠,根据随机原则分为 2 组:正常组和糖尿病模型组。糖尿病造模成功后不进行治疗,高脂饲料喂养 2 周后,将糖尿病大鼠随机均分为糖尿病组、脑梗塞假手术组、模型组、治疗组 4 组。造模完成后分别灌胃,治疗组每日以复元醒脑汤灌胃,正常组、糖尿病组、假手术组、模型组予等容量的生理盐水灌胃,每天 2 次,连续 3~7 d。每天对其恢复程度进行神经体征评分;末次灌胃后,分别检测各组大鼠血糖、血清胰岛素、稳态葡萄糖输注率等指标。结果 模型组与假手术组、糖尿病组、正常组比较,神经缺损行为体征评分升高(P<0.01),胰岛素抵抗程度加重(P<0.01);与模型组相比,治疗组神经体征缺损明显改善(P<0.01)、胰岛素抵抗减轻。结论 (1)本实验在 SD 大鼠糖尿病的基础上采用自体血栓法建立糖尿病脑梗塞模型。该模型是与人类原发脑梗塞的机理较为接近的动物模型;(2)糖尿病脑梗塞大鼠出现胰岛素抵抗加重等现象;(3)复元醒脑汤对糖尿病脑梗塞大鼠术后神经体征缺损具有明显改善作用,并减轻了糖尿病脑梗塞大鼠体内胰岛素抵抗程度。

Objective we are going to carry out the experimental study on"Fuyuan Xingnao Decoction"in treating Cerebral infarction,in order to offer theoretical and experimental foundation. Methods SPF SD male rats were randomly divided into 2 groups:normal group, diabetic model group. Diabetes mellitus after injecting untreated, high fat diet for 2 weeks later, the diabetic rats were randomly divided into diabetic cerebral infarction group, sham operation group, model group, treatment group 4 group. Mold making respectively after gastric lavage, the treatment group of Fuyuan Xingnao Decoction daily by gavage, normal control group, diabetic group, sham operation group,model group to wait for volumes of saline lavage,2 times a day, continuous 3-7 d. Record the degree of recovery of neurological signs score everyday;the last after intragastric administration of glucose, were used to detect the rats, serum insulin, steady-state glucose infusion rate. Results Compared with sham operation group, diabetic group, normal group, the model group cerebral neurological behavior symptoms scores decreased (P〈 0.01 ), the degree of insulin resistance worsen(P〈 0.01). The treatment group compared with the model group, significantly improved the neurological signs defects,insulin resistance loss. Conclusions 1. The experiment using autologous clot method on the basis of SD rat diabetes to establish the model of diabetic cerebral infarction. The model is associated with human primary brain infarction mechanism relatively. 2. Diabetes mellitus cerebral infarction rat were found that the aggravation of insulin resistance increased. 3. "Fuyuan Xingnao Decoction" on diabetic cerebral infarction rat postoperative neurological signs defect has significantly improve, and the reduction of the diabetic cerebral infarction rats in insulin resistance degree.
