用显微技术观察了贻贝棘尾虫对碳纳米管(CNTs)的摄取以及虫体分裂时CNTs在其体内的分布,测定了0.1、0.5、1.0、5.0、10、50和200μg·mL^-1 7个浓度的CNTs与贻贝棘尾虫存活率的响应曲线,发现加CNTs后第5天虫体的存活率分别为111%、107%、75%、70%、53%、43%、38%左右,表明高浓度的CNTs对细胞的生长有与浓度相关的抑制作用,而当浓度降至足够低时。CNTs对细胞的生长有促进作用。在此基础上用PI染色的方法初步探讨了高浓度CNTs对细胞生长产生抑制作用的原因,荧光显微镜观察结果显示CNTs损伤了贻贝棘尾虫大核和膜的正常生理功能。
The uptake of CNTs by Stylonychia mytilus and their distribution during the dividing process of the cells were observed with optical microscope for the first time. The dose-effect relationship between concentration of CNTs and surviving rate of Stylonychia mytilus was measured. The surviving rotes were found to be about 111%, 107%, 75%, 70%, 53%, 43%, 38% on the fifth day after adding CNTs for the concentrations of 0.1, 0.5, 1, 5, 10, 50, 200 μg·mL^-1, respectively. The result showed that high concentration of CNTs exhibited a dose-dependent inhibition to the cells, whereas very low concentration of CNTs was likely to stimulate the growth of the cells. To understand the inhibition from the CNTs, the propidium iodide was used to dye the macronucleus of Stylonychia mytilus. Images from fluorescent microscope showed that the damage occurred on the macronucleus and membrane of the cells.
