黄连解毒汤抗铜绿假单胞菌生物被膜及与阿奇霉素协同抗菌作用

目的:比较益气活血汤、镇肝熄风汤和星蒌承气汤3种中药复方对急性脑缺血再灌注大鼠缺血脑组织Glu含量与NR1表达的影响。方法:采用线栓法阻断大鼠左侧大脑中动脉制作局灶性脑缺血再灌注模型。大鼠随机分为假手术组、模型组、镇肝熄风汤和星蒌承气汤组。采用高效液相色谱法和免疫组化SABC染色法,分别检测Glu的含量和NR1表达。结果:脑缺血2h再灌注各时间点与假手术组比较,模型组Glu含量和NRI表达均显著升高和增加(P〈0.05或P〈0.01);与模型组比较,各治疗组各时间点Glu含量和NR1表达显著降低和减少(P〈0.01);治疗组组间比较,48h无显著差异,6h星萎承气汤组在减少NRl表达方面最优(P〈0.05);结论:两种中药复方可以通过降低Glu含量和减少NR1表达,减轻兴奋性氨基酸毒性,抗脑缺血再灌注损伤。

英文摘要:

Objective: To compare the effects of two traditional Chinese medicinal compounds on content of glutamate and expres- sions of NR1 in cerebral tissue of rats after focal cerebral ischemia and reperfusion (I/R) . Methods: The local cerebral I/R model was established by ligation of the middle cerebral arteries (MCA) . The animals were divided into the sham - operative group, the model group, the Zhengan Xifeng Decoction (ZXD) group and the Xinglou Chengqi Decoction (XCD) group. The contents of gluta- mate was detected by HPLC. Im -munohistochemical SABC method was used to obsevre the expressions of NR1. Rdusts: Compared with-those of the sham- operative group, the contents of glutamate and expressions of NR1 increased markedly at all time pionts after I/R in model group. Compared with those of model group, the contents of glutamate and expressions of NR1 decreased or reduced at all time pionts after I/R in all treatment groups. The optimal effects were shown in XCD at 6 h in reducing expressions of NR1. Con- clusion: The two traditional Chinese medicinal compounds could reduce pathologic injury after focal cerebral I/R by decreasing or re- ducing contents of glutamate and expressions of NR1.
