慢性牙周炎和动脉粥样硬化动物模型的建立及研究

目的应用Wistar大鼠建立慢性牙周炎和动脉粥样硬化动物模型,探讨与慢性牙周炎和冠心病发病相关的可能机制。方法将60只雄性Wistar大鼠按随机数字表分为4组:A组:正常对照组,B组:慢性牙周炎组,C组:动脉粥样硬化组,D组:慢性牙周炎+动脉粥样硬化组,每组15只。各组分别按照实验设计进行相应干预处理,至12周末处死动物,分别检查记录牙周指标,检测血清中的总胆固醇、低密度脂蛋白水平、肿瘤坏死因子α(TNF-α)以及基质金属蛋白酶3(matrix metalloproteinase,MMP-3)的含量,进行统计学分析,并观察牙周和腹主动脉的组织病理学改变。结果经过实验牙牙周检查、血清学检测以及组织病理学观察,已成功建立了慢性牙周炎及动脉粥样硬化动物模型。牙周病理结果显示,B、D组实验牙出现明显牙周炎症,其附着丧失[B、D组分别为(137.86±28.39)、(162.36±22.69)μm]较A、C组[分别为(4.26±1.07)、(68.07±18.25)μm]显著增加(P〈0.05),C组附着丧失较A组也有显著增加(P〈0.05)。C、D组腹主动脉病理学观察显示形成明显粥样硬化病变。C、D组总胆固醇[分别为(6.69±0.69)、(9.17±1.82)mmol/L]及低密度脂蛋白水平[分别为(5.58±0.52)、(6.29±0.72)mmol/L]明显高于A[分别为(0.68±0.34)、(0.26±0.09)mmol/L]、B组[分别为(1.17±0.26)、(0.69±0.19)mmol/L],P〈0.05;D组总胆固醇及低密度脂蛋白水平显著高于C组(P〈0.05)。血清TNF-α、MMP-3水平B、D组显著高于A、C组(P〈0.05)。D组血清MMP-3与总胆固醇(P=0.971)、低密度脂蛋白(P=0.604)之间无明显相关性。结论慢性牙周炎与动脉粥样硬化可能具有一定的相关性,可能对动脉粥样硬化和冠心病的发生、发展有重要的影响;MMP-3可能是独立于血脂之外的?

Objective To investigate the possible correlation between atherosclerosis and chronic periodontitis by establishing an animal model of chronic periodontitis and atherosclerosis in Wistar rat. Methods Sixty male Wistar rats were divided into four groups: A (control group), B (chronic periodontitis group), C ( atherosclerosis group), D ( chronic periodontitis accompany with atherosclerosis group). Every group was accepted the corresponding treatment. Animals were sacrificed after 12 weeks. The periodontal index, levels of serum total cholesterol (TC) and low-density lipoprotein (LDL), the concentration of TNF-α and matrix metalloproteinase (MMP-3) were examined. The severity of chronic periodontitis and atherosclerosis was quantified by histopathology. The date were statistically analyzed. Results Through detection of periodontal tissue of experimental teeth, serum and histopathology, animal models were successful. Histopathologic observation revealed : obvious inflammation of periodontal tissue was observed in group B and D. Attachment loss level in group B[ (137.86 ±28.39) μm] and D [ (162. 36 ±22. 69) μm] was higher than that in group A[ (4. 26 ± 1.07) μm ] and C[ (68.07 ± 18.25) μm ] (P 〈 0. 05 ), and that in group C was higher than group A( P 〈 0. 05 ). Atherosclerotic lesions of abdominal aorta were formed in group C and D. The level of TC, LDL in group C and D was higher than that in group A and B ( P 〈 0. 05 ), and that in group D was higher than group C ( P 〈 0. 05 ). Animals in group B and D showed higher level of TNF-α, MMP-3 in serum than that in group A and C ( P 〈 0. 05 ). There was no correlation between the level of MMP-3 and TC (P = 0. 971 ) or LDL (P = 0. 604). Conclusions Chronic periodontitis may be a risk factor and contribute to the pathogenesis of atherosclerosis. MMP-3 may be an independent risk factor of atherosclerosis exclude TC and LDL.
