目的观察原代培养大鼠皮质神经元生长过程中β-Tubulin的表达变化并进行图像分析。方法取孕13-14天胚鼠大脑皮质接种于覆有多聚赖氨酸的爬片上,培养液为添加了B27(v/v%:2%)和Glutamax(v/v%:1%)的Neurobasl Medium(Gibco)。细胞爬片进行免疫细胞化学染色,显微镜观察,LeicaDMR+0550图像分析系统进行图像分析。结果β-Tubulin表达和分布随着神经元发育成熟而变化,生长初期主要分布于核周附近,随神经元的成熟散在分布于胞质及突起内,构成细胞骨架,维持细胞形态。结论在神经元生长过程中伴随有β-Tubulin的表达变化,随神经元的成熟而构成细胞骨架,维持细胞形态。
Objective To observe the expression changes of β-Tubulin in growth process of primarily cultured rat cortical neurons and analyze the corresponding images. Methods Embryonic cortical neurons of 13-14 day pregnant embyo SD rat were cultured in slide coated by poly-lysine with serum- free medium containing B27 (v/v % 2 %) and Glutamax(v/v %: 1 % ). Immunocytochemistrical staining were used to observe neurons with OLYMPUS BX60 microscopy and LeicaD MR+Q550 image analysis system. Results During the different phases of the muturation process in the rat cortical neurons there were expression changes of β-Tubulin : in the early phase it showed an obvious distribution around nuclei, and then, with the maturation of neurons, it was scattered in cytoplasm to compose the cell skeleton and sustain the cell shape. Conclusion Expression changes of Tubulin are concomitant with the different phases of growth process in neurons, and, with the maturation of neurons, it composes the cell skeleton and sustain the cell shape.
