目的:探讨四妙勇安汤干预动脉粥样硬化(AS)易损斑块内细胞外基质(ECM)的作用机制。方法:随机将40只日本大耳白兔分为正常组、模型组、辛伐组、四妙组,第24周取材。检测第8、16、24周时血清基质金属蛋白酶-9(MMP-9)含量;检测动脉壁胶原含量(CA)、细胞外脂质含量(ELA)、脂核与斑块面积比(LCA/PA)及纤维帽厚度(FCT);免疫组化方法检测斑块内MMP-9、α-平滑肌肌动蛋白(α-SMA)及核因子-κB(NF-κB)阳性面积百分比。结果:给药后各时间点四妙组血清MMP-9水平均低于模型组(P〈0.01);斑块内ELA、LCA/PA、MMP-9和NF-κB阳性面积百分比均低于模型组,斑块FCT、CA及α-SMA阳性面积百分比均高于模型组(P〈0.05或P〈0.01)。结论:四妙勇安汤能够抑制细胞外基质降解,稳定AS易损斑块,其作用机制可能是通过抑制MMP-9表达实现的。
Objective:To observe the intervening effect and acting mechanism of Simiaoyongan Decoction(SYD) on extracellular matrix in rabbit's Atherosclerosis plaque.Methods:40 Japanese white rabbits were randomly assigned to two groups:10 animals in the normal control group,and 30 animals in experiment group which was established to the AS model by high fatty diet feeding,combined with immune injury and femoral arterial balloon tearing.At the 8th week,30 animals in experiment group were randomly assigned to three groups:model group,SYD group and simvastatin group in which there are 10 animals.The medication of SYD and simvastatin was given in the SYD group and simvastatin group.MMP-9 were measured at the 8th,16th and 24th week.Levels of Collagen,α-SMA,MMP-9 and NF-κB expression in aortic wall were measured at the terminal of experiment.Results:Compared with the model group,the level of MMP-9 in SYD group decreased at various time points;MMP-9 and NF-κB expression in aortic wall were lower in the treated groups after treatment;Collagen and α-SMA level increased(P0.01or P0.05).Conclusion:SYD could suppress degradation of extracellular matrix to stabilize atherosclerosis vulnerable plaque.Its mechanism can be the restraint in MMP-9 and a cell signaling protein NF-κB expressions
