目的应用呼吸曲线Penh值评估哮喘模型大鼠的气道高反应性,以探讨一种无创性检测哮喘大鼠气道高反应性的方法。方法采用卵清白蛋白腹腔内注射致敏和雾化吸入激发的方法制备大鼠哮喘模型,用整体容积描记箱(体描箱)无创性记录呼吸曲线,推算Penh值并观察乙酰甲胆碱(Mch)刺激后的变化,然后用传统有创性气管插管的方法测定气道反应性,行肺组织病理切片染色。结果哮喘模型大鼠表现出气道高反应性,Mch刺激后呼吸频率和Penh值明显增加,根据Penh值的变化计算出的Mch负对数浓度和传统方法测定的气道反应性的定量指标呈明显正相关。结论体描箱呼吸曲线能够反应哮喘大鼠气道反应性的变化,可成为一种无创性检测哮喘大鼠气道高反应性的方法。
Objective To explore a noninvasive method for measuring airway hyperreactivity in asthma rats. Methods The asthma models of Wistar rats were established by OVA intraperitoneal injection and inhalation. Whole-body plethysmography was used to record the change after Mch challenge, then tracheas were intubated for traditional measure of airway reactivity. Results Respiratory rate and Penh were obviously increased after challenge in asthma rats. The negative log concentration at 50% increase of Penh was closely related to that of peak airway pressure at 50% increase. Conclusion Pneumotachography by whole-body plethysmography is prospective for noninvasive measurement of airway hyperreactivity in asthma rats.
