目的考察延胡索在大鼠血浆中的药动学特征及炮制对其吸收和代谢的影响。方法采用高效液相测定灌胃延胡索提取物后的大鼠血浆中的药物浓度。结果建立了同时测定大鼠血浆中延胡索乙素,原阿片碱和去氢紫堇碱的方法,该方法的低,中,高3种药物浓度的回收率,日内和日间精密度均符合要求。结论醋炙能加快延胡索乙素和去氢紫堇碱在体内的吸收,同时延缓二者的消除,其中对延胡索乙素的作用更明显,而对原阿片碱影响不大;酒炙对延胡索乙素的作用与醋炙相似,但作用较醋炙弱,同时对原阿片碱和去氢紫堇碱均有延缓消除的作用。
OBJECTIVE To investigate the pharmacokinetics of Rhzoma Corydalis in rat plasma and the influence of the decoction process on its absorption and metabolism. METHODS The blood drug level in rats after oral administration of Rhzoma Corydalis extraction was determined by HPLC. RESULTS The method for simultaneous determination of tetrahydropalmatine, protopine and dehydrocorydaline of Rhzoma Corydalis in rat plasma was established by HPLC. The intra-day and inter-day validation with low, middle, high concentrations were good. CONCLUSION The results of samples analysis showed that decocted products with vinegar can promote absorption and delay elimination of tetrahydropalmatine and dehydrocorydaline in vivo. The deeocted products with liguor can delay elimination of tetrahydropalmatine, protopine and dehydrocorydaline in vivo. The influence of the process on tetrahydropalmatine is more conspicuous than protopine and dehydrocorydaline.
