目的:探讨下丘脑瘦素受体(leptin receptor,Ob-R)改变与多囊卵巢综合征(Polycystic ovary syndrome,PCOS)发病机制的关系。方法:将45只雌性SD大鼠分为正常对照组、溶剂对照组和实验组。溶剂对照组用1%羟甲基纤维素水溶液(CMC)灌胃,实验组用来曲唑制造PCOS大鼠模型,比较各组大鼠体重、动情周期、卵巢重量及卵巢形态学的改变,测定血清黄体生成素(LH)、睾酮(T)、胰岛素(INS)、空腹血糖(FG)、瘦素(LEP)水平,用免疫组织化学和Real—TimeRT—PCR方法检测大鼠下丘脑Ob—R及mRNA的相对表达。结果:PCOS大鼠血清T、LH、INS、LEP水平明显高于正常对照组和溶剂对照组(P〈0.05),PCOS大鼠下丘脑瘦素受体表达较正常对照组和溶剂对照组明显减弱(P〈0.05)。结论:瘦素受体参与PCOS大鼠内分泌紊乱的发生,下丘脑瘦素受体表达的下调可能是PCOS大鼠瘦素抵抗的原因之一。
Objective:To study the relationship between leptin receptor(Ob-R) expression on the hypothalamus and the pathogenesis of the polycystic ovary syndrome (PCOS). Methods:Forty five female SD rats were divided into three groups:normal control group, solvent control group and model group( PCOS group). The body weight ,estrous cycle ,the ovary weight and ovary morphological changes were observed and compared. Serum LH, T, INS, FG, LEP levels were measured. The relative expression of Ob-R on the hypothalamus of each group was detected by immunohistoehemistry and real time RT-PCR. Results:Serum testosterone, luteinizing hormone, insulin,leptin levels were higher in PCOS group than those of the normal control group and the solvent control group (P 〈 0.05 ). The expression of Ob-R on the hypothalamus of the PCOS group was weaker than that of the normal control group and the solvent control group (P 〈 0.05 ). Conclusion :The leptin receptor takes part in the occurrence of the endocrine disturbance in PCOS rats. The decrease expression of leptin receptor on the hypothalamus of the PCOS rats may be one of the reasons of the leptin resistence in the PCOS rats.
