目的:探讨不同强度的运动对糖尿病动脉粥样硬化大鼠主动脉组织Toll样受体2(TLR-2)、Toll样受体4(TLR-4)的影响.方法:选取健康SD大鼠100只,随机分成对照组20只及糖尿病动脉粥样硬化组80只,糖尿病动脉粥样硬化模型采用高糖高脂饮食加链脲佐菌素(STZ)(30 mg/kg)注射建立.其中糖尿病动脉粥样硬化组又分成模型组、小运动量组、中等运动量组和大运动量组(每组20只).经过估计力竭运动时间及游泳训练后,小运动量组(12 min/次),中等运动量组(21 min/次),大运动量组(30 min/次),进行6周的运动干预,1次/d,一周6次.用PCR法测定各组大鼠主动脉组织的TLR2、TLR4mRNA的表达.结果:模型组较对照组血管组织中TLR2、TLR4水平升高;运动干预后,各运动组TLR2、TLR4水平较模型组降低,以中等运动量、大运动量组下降明显.结论:糖尿病动脉粥样硬化大鼠主动脉组织中TLR2、TLR4水平升高,TLR2、TLR4水平的升高可能是糖尿病动脉粥样硬化的发病机制之一,运动能够降低TLR2、TLR4的表达.
Objective:To study the effect of different intensity of exercise training on the expression of TLR2 and TLR4 in diabetic atherosclerosis rat aorta.Methods:One-hundred Sprague-Dawley rats were randomly assigned into one of five groups:control group (CG,n =20) and diabetes atherosclerosis group(DAG,n =80),After given high-fat-high-sugar diet plus streptozotocin injection (30 mg/kg) for 4 weeks,the diabetes atherosclerosis group was divided into four subgroups:the model group(MG,n =20),the low intensity exercise group(LEG,n =20),the moderate intensity exercise group(MEG,n =20) and the high intensity exercise group (HEG,n =20).According to the exhausting swimming time,the exact time of HEG / MEG/ LEG in this study were 30 minutes,21 minutes,12 minutes.The rats were sacrificed after 6-week swimming,and the expressions of TLR2 and TLR4mRNA in aorta tissue of rats was tested by reverse transcriptase polymerase chain reaction (RT-PCR).Results:Compared with the control group,the concentration of TLR2 and TLR4 in model group was higher in blood vessels; After the intervention of exercise,The concentration of TLR2 and TLR4 in every exercise group decreased obviously,especially in the moderate exercise group and the great physiological load of exercise group.Conclusion:The concentration of TLR2 and TLR4 in aorta tissue of diabetes atherosclerosis rat will increase.The increased TLR2 and TLR4 could be one of the pathogenesis of atherosclerosis in diabetes,exercise can reduce the expression of TLR2 and TLR4.
