目的观察细粒棘球蚴囊液及原头蚴对体外培养BABL/c小鼠脾细胞TGF-β表达的影响。方法取BABL/c小鼠脾细胞,制成细胞悬液,加入不同浓度囊液/原头蚴共培养72h,用ELISA检测上清液TGF-β含量;同时收集细胞并提取mRNA,用RT-PCR检测TGF-βmRNA的相对表达量。试验以PBS与脾细胞混合培养组为对照。结果 ELISA检测囊液组和原头蚴组TGF-β分泌均增加,与对照组相比差异有统计学意义(P〈0.05);RT-PCR检测囊液组和原头蚴不同浓度组及对照组TGF-βmRNA的相对表达量,差异均有统计学意义(P〈0.05),但囊液/原头蚴不同浓度组间差异无统计学意义(P〉0.05)。结论原头蚴和囊液均能刺激体外培养小鼠脾细胞TGF-β表达增加,提示原头蚴和囊液中含有能促使宿主产生TGF-β的抗原物质,这些抗原物质在包虫病的免疫逃避中发挥一定作用。
Objective To observe the effect of treatment with Echinococcus granulosus fluid/protoscoleces on TGF-β expression in mouse spleen cells in vitro. Methods After spleen cells from BABL/c mice were co-cultured with different concentrations of hydatid fluid or protoscoleces, the level of TGF-β in the co-culture supernatant were measured using ELISA. mRNA extracted from cells was collected and the relative level of expression of TGF-β mRNA was detected using RT-PCR. Spleen cells were co-cultured with PBS to serve as a control. Results ELISA indicated that, compared to the controls, cells co-cultured with hydatid fluid and protoscoleces had increased secretion of TGF-β; the difference in secretion was statistically significant (P〈0.05). RT-PCR to detect the relative level of expression of TGF-βmRNA indicated that, compared to the controls, cells co-cultured with hydatid fluid and protoscoleces had significantly increased expression (P〈0.05), but this increased expression did not differ significantly with different concentrations of hydatid fluid or protoscoleces. Conclusion The secretion of TGF-β increased in mouse spleen cells treated with hydatid fluid and protoscoleces in vitro, indicating that hydatid fluid and protoscoleces may contain some antigenic substance that induces the host to produce TGF-β, facilitating evasion of the host's immune system.
