目的:研究翻白草乙醇提取物对人肝癌细胞HepG-2体外增殖的抑制作用及凋亡的诱导作用。方法:用不同浓度的翻白草乙醇提取物对人肝癌细胞HepG-2,MTT法测定对细胞生长的抑制率,用琼脂糖凝胶电泳观察DNA梯状条带和DAPI染色观察凋亡小体,再用Western blot法检测对caspase-3和PARP蛋白表达的影响。结果:翻白草乙醇提取物对人肝癌细胞HepG-2有较强的浓度依赖性抑制作用,诱导细胞出现明显的形态学改变,琼脂糖凝胶电泳出现典型的DNA梯状条带,Western blot分析结果发现,翻白草乙醇提取物促进caspase-3活化而引发下游PARP裂解,进而诱导人肝癌细胞HepG-2细胞走向自体凋亡的途径。结论:翻白草乙醇提取物可能通过抑制人肝癌细胞HepG-2的增殖及诱导其凋亡而产生抗肿瘤作用。
AIM:Potentilla discolor Bunge (PD),Chinese traditional medicinal herb,was investigated for its antitumor activity and induction of apoptosis in vitro,using human HepG-2 hepatocellular carcinoma cell line.METHODS:The viability of HepG-2 cells was evaluated by MTT,after incubation with ethanolic extract of PD (5-200 mg L-1).And HepG-2 cells were manipulated by fluorescent microscopic analysis stained with DNA-specific fluorochrome 4',6-diamidino-2-phenylindole dihydrochloride and performed DNA fragmentation analysis.And caspase-3 activity and poly (ADP-ribose) polymerase (PARP) cleavage were detected by western blot.RESULTS:PD decreased the cell viability in a concentration-dependent manner and exhibited typical characteristics of apoptosis.Active form of caspase-3 and cleaved PARP were found in concentration-dependent manner.CONCLUSION:These results indicated that PD may represent a potential new source of anticancer candidate.
