目的 观察人类主要细胞周期素(Cyclin D1、E、A、B1)在结直肠癌中非时相表达特征,探讨主要细胞周期素表达分型与结直肠癌生物学行为的关系.方法 用流式细胞术(FCM)双参数法半定量检测83例结直肠癌手术切除标本细胞中Cyclin D1、E、A、B1表达.根据不同Cyclin在细胞周期中的作用及其异常表达特征,将其表达分为不同类型及亚型结合临床病理学资料进行分析.结果 结直肠癌细胞Cyclin(D1、E、A、B1)异常表达大致可归为5种类型:Ⅰ型Cyclin(D1、E、A、B1)均高表达;Ⅱ型Cyclin D1低表达,Cyclin(E、A、B1)均不同形式高表达;Ⅲ型Cyclin D1不表达,Cyclin(E、A、B1)均不同形式高表达;Ⅳ型Cyclin D1不表达,Cyclin(E、A、B1)不同形式低表达;Ⅴ型Cyclin(D1、E、A、B1)均不表达;Ⅱ型→Ⅳ型根据不同周期内Cyclin的优势表达分为相应不同的亚型.a亚型:Cyclin(E、B1)表达为主;b亚型:Cyclin E表达为主;c亚型:Cyclin B1表达为主;d亚型:Cyclin(E、A、B1)表达为主.主要Cyclins表达类型与结直肠癌淋巴结转移率、转移程度、浸润程度明显相关(P<0.01).主要Cyclins表达类型Ⅰ→Ⅴ型结直肠癌淋巴结转移率、转移程度、浸润程度呈上升趋势.结直肠癌的生长方式和细胞分化程度与主要Cyclins表达类型的亚型明显相关(P<0.01).结论 主要Cyclins(D1、E、A、B1)表达类型与结直肠癌生物学行为有关,可以作为一种体现结直肠癌生物学行为较为客观的分类方法.
Objective To investigate the expression characters of human primary Cyclins in different colorectal adenocarcinomas and explore the relationship between the types of primary Cyclins expressed and biological behavior of colorectal adenocarcinomas. Methods Primary Cyclins ( Dl, E, A, B1) expressed in single cells detached from 83 cases of colorectal adenocarcinomas tissues were analyzed by flow cytometry (FCM). Colorectal adenocarcinomas were classified by different types of primary Cyclins expressed or sub-type according to the roles of primary Cyclins expressed in cell cycle and the expression patterns of Cyclins. The results were analyzed in combination with clinical pathomorphology of carcinomas. Results The expression patterns of primary Cyclins could be classified five types: type I : high expression of Cyclin D1 and Cyclins (E, A, B1); type II : low expression of Cyclin D1, high expression of Cyclins (E, A, B1) by different patterns; type HI: no expression of Cyclin D1, high expression of Cyclins (E, A, B1) by different patterns; type IV : no expression of Cyclin Dl, low expression of Cyclins (E, A, Bl) by different patterns; type V : no expression of in-cycle Cyclins or out-cycle Cyclin. Each type could be, according to the degree of in-cycle Cyclins expressed, divided into different sub-types. The types of primary Cyclins expressed were strongly linked to invasive depth and lymph node metastasis of colorectal adenocarcinomas (P 〈0.01). The degree of invasion, and the degree and the rate of lymph node metastasis were increased gradually from type I to type V. The type of primary Cyclins expressed was also significantly associated with disease stage (TNM stage). The proportion of stage IV disease was increased gradually from type I to type V. It was shown there were relationships between the sub-types of primary Cyclins expressed and different growth-types or degrees of cell differentiation in colorectal adenocarcinomas (P 〈0.01).
