目的观察麻杏石甘汤对流感病毒感染小鼠肺部炎症与超微结构的影响。方法采用鼻腔接种法建立流感病毒小鼠肺部感染模型。实验设正常对照组、模型对照组、利巴韦林组和麻杏石甘汤组,经灌胃给药或给生理盐水7 d后,处理动物,常规方法检测肺重量指数;光学显微镜技术观察肺病理变化;透射电子显微镜技术观察肺组织超微结构。结果①与正常对照组比较,模型对照组小鼠肺指数明显升高(P〈0.05),麻杏石甘汤组小鼠肺指数降低,与模型对照组比较,差异有统计学意义(P〈0.05);②与正常对照组比较,模型对照组小鼠肺脏出现明显的病理变化,肺泡、细支气管等正常组织结构消失,肺泡内有大量炎症细胞浸润,麻杏石甘汤组小鼠肺组织病理变化显著改善,偶可见少量炎症细胞浸润;③与正常对照组比较,模型对照组小鼠肺组织超微结构受到破坏,核膜与质膜模糊不清,细胞浆内细胞器减少,出现空泡样病变,发现病毒颗粒,麻杏石甘汤组小鼠肺组织的细胞超微结构较完整,核膜与质膜清晰可见,细胞器比模型对照组丰富,细胞浆内空泡样病变程度明显低于模型对照组。结论麻杏石甘汤对A型流感病毒感染所致的肺部炎症与超微结构有改善作用。
【Objective】Observing the effect of Maxing Shigan Decoction(MXSGD) to inflammation and ultrastructure in lungs of mice infected by influenza virus.【Methods】Using nasal inoculation to establish mouse pulmonary infection model animals by A-type influenza virus.Set up normal group,model group,ribavirin group,MXSGD group.After 7 days of lavaging medicine or normal saline,disposed mice and detected pulmonary weight index routine;Observing pulmonary pathologic changes and recorded the pathologic property and degree by light microscope;Observing pulmonary ultrastructure by transmission electron microscope.【Results】Compared with normal group,pulmonary index of model group is significant go up(P 0.05).Pulmonary index of MXSGD group go down,which different from model group observably(P 0.05).Compared with normal group,lungs of model group are hyperemic and dropsical obviously,while those normal organizations and structures such as alveolus,bronchioles were disappeared,mass inflammatory corpuscle infiltrate within lungs.Pulmonary pathologic changes were significant improving in MXSGD group.Compared with normal group,pulmonary organization structures are destroyed in model group,karyolemma and karyoplasm were inexplicit,the quantity of organelles were decreased,vacuolated changes showed up and there were plenty of viral particles in endochylema.In MXSGD group,pulmonary organizational structure nearly intact,karyolemma and karyoplasm were clearly visible,organelles were plentiful pampered with control group,degree of vacuolated changes within endochylema was lower than model group.【Conclusion】MXSGD play a part in mending inflammation and ultrastructure in lungs of mice infected by influenza virus.
