目的探讨脐带血与成人血波尔效应及协同效应的差异。方法收集新鲜脐带血与成人血各3份,用氧平衡曲线仪,按先检测氧离曲线,加10μl 1mol/LHCl后继续检测氧离曲线,加15μl1mol/LNaOH后再次检测氧合曲线的顺序.检测各样本的氧离(合)曲线,并读取各样本氧离(合)曲线的P50与希尔系数(HC)。比较两组样本P50的变化值(APS0)与变化率(%AP50)。根据各组均值用希尔方程绘制曲线比较移动幅度。结果在HCl及NaOH作用下,脐带血P50、HCHCl、HCNaOH、△P50HCl、%△P50HCl、△P50NaOH、%△P50NeOH均显著低于成人血(P〈0.05),氧离(合)曲线移动幅度不及成人血。结论在酸(碱)环境下,脐带血氧亲和力高于成人血,而波尔效应及协同效应均差于成人血。
Objective To explore the difference of Bohr effects and synergistic effects between umbilical cord blood and adult peripheral blood. Methods Three samples of umbilical cord blood and adult peripheral blood were collected respectively. The oxygen dissociation curve (ODC) was detected firstly with oxygen balance curvometer, and then, 10μl 1 mol/L HCl was added and the ODC was detected; next, 15μl 1 mol/L NaOH was added to detect the oxygenation curve (OGC), and the ODC and OGC in each sample were detected and PS0 and hill coefficient (HC) of such curved were read. The change (AP50) and rate of change (%△P50) of P50 of the samples of umbilical cord blood and adult peripheral blood were compared. Curves were drawn with Hill equation and based on the mean of the two groups. Results Under the effects of HCl and NaOH, the P50, HCHa, HCNaOH, △P50HCl, %△P5HCl, AP50Naos, %△P50NaOH of umbilical cord blood were significantly lower than those of adult peripheral blood(P 〈 0.05). The shift of ODC and OGC of umbilical cord blood was less than that of adult peripheral blood. Conclusion In acid (alkali) environment, the oxygen affinity of umbilical cord blood is higher than that of adult peripheral blood. However, the Bohr effects and synergistic effects of umbilical cord blood are inferior to those of adult peripheral blood.
