p53凋亡刺激蛋白2(ASPP2)是ASPP家族的一个重要成员,能够通过对细胞促凋亡靶基因转录的调节来促进细胞的凋亡。另外,ASPP2与自噬也具有一定的关系,它能通过提高p53通路介导的自噬来诱发肿瘤细胞凋亡增加,从而达到抑制和杀灭肿瘤的作用。在肝癌细胞中,ASPP2过表达可增强p53诱导的自噬调节蛋白(DRAM)介导的自噬对肝癌细胞的促凋亡作用。DRAM的发现使p53与自噬之间建立了联系,也进一步丰富了ASPP2的功能机制,这也为肿瘤的基因治疗提供了新的思路。
Apoptosis stimulating protein 2 of p53 (ASPP2)is an important member of apoptosis stimulating protein. As a cofactor of p53 family, it can activate and transcribe the apoptosis stimulating gene, and promote cell apoptosis. In addition to, ASPP2 could enhance the autophagy mediated by p53 pathway to induce the increasing of the apoptosis of tumors and depress or kill the tumor. The over expression of ASPP2 could enhance autophagy mediated by DRAM in the apoptosis of HCC cells. DRAM is the first p53 target protein reported to induce autophagy, which establishes contact between p53 and autophagy, and enriches the functional mechanisms of ASPP2, providing a new approach for gene therapy in some tumors.
