目的检测脯氨酸羟化酶(proline hydroxylase domain 2, PHD2)在结直肠癌及癌旁肠黏膜中的表达,探讨分析其临床意义。方法应用组织芯片技术结合免疫组化法检测本校3所附属医院收治的结直肠癌患者术后149例组织标本以及相应50例癌旁肠黏膜和50例结直肠癌原发及转移灶中PHD2 蛋白的表达。结果PHD2在149例结直肠癌组织中阳性表达率显著低于癌旁肠黏膜(48.9% vs 72.0%, P〈0.01),并与肿瘤的分化程度、临床分期、转移等密切相关(P〈0.01);在50例结直肠癌原发灶中阳性表达率高于相应转移灶(64.0% vs 46.0%, P〈0.01)。结论PHD2在结直肠癌组织中表达低于癌旁肠黏膜,在结直肠癌原发灶中表达高于相应转移灶,在结直肠癌的恶性进展中发挥着重要作用。
Objective To investigate the expression status of proline hydroxylase domain 2 (PHD2) in colorectal cancer and adjacent tissue, and to explore the clinical significance of PHD2.Methods Immunohistochemical staining was applied for the detection of PHD2 expression in 149 cases of colorectal cancer tissues, 50 adjacent mucosa tissues and 50 cases of metastasis from three affiliated hospitals.Results The expression level of PHD2 in the colorectal cancer tissue was lower than that in the adjacent tissue. The positive expression rate of PHD2 in the colorectal cancer tissue was significantly lower than that in the adjacent tissue (48.9% vs 72.0%, P〈0.01). PHD2 expression was significantly correlated with tumor differentiation degree, clinical stage and metastasis (P〈0.01). The expression level of PHD2 in the colorectal cancer tissues was higher than that in the metastasis (64.0% vs 46.0%, P〈0.05).Conclusion PHD2, which is mildly expressed in colorectal cancer tissue but highly expressed in adjacent tissue, plays an important role in the malignant progression of colorectal cancer.
