目的:探讨EphB4/ephfinB2信号通路在血府逐瘀汤促血管新生中的作用。方法:运用血清药理学方法,以1.25%、2.5%、5%浓度的血府逐瘀汤含药血清和空白血清干预人脐静脉内皮细胞(Human Umbilical Vein Endothelial Cells.HUVEC-12)48h,通过MTT法、Boyden小室迁移法、细胞黏附法、逆转录PCR法分别检测细胞增殖、迁移、黏附、EphB4和EphrinB2基因表达的情况。结果:与同血清含量的对照组比较,5%浓度含药血清对细胞的增殖能力起抑制作用。1.25%、2.5%和5%3个浓度含药血清皆有显著的促内皮细胞迁移和黏附作用,并能抑制EphB4的表达,其中1.25%、2.5%浓度含药血清抑制EphfinB2的表达。结论:血府逐瘀汤通过促内皮细胞迁移和黏附发挥促血管新年作胃.日该作用可能与EphB4/EphrinB2通路密切相关。
object To explore the roles of EphB4/ephrinB2 in Angiogenesis induced by Xuefu Zhuyu Decoction (XZD). Methods: Using serum pharmacology technique to study the cell proliferation, migration, adhesion, ephrinB2 and EphB4 gene expression by MTT, Boyden chamber, ceil adhesion and reverse transcriptase polymerase chain reaction( RT- PCR) separately on Human Umbilical Vein Endothelial Cells (HUVEC-12) after intervened by 1.25% , 2.5% and 5% XZD containing serums for 48h. Results Comparison with the control group of same serum level , 5% medicated serum inhibited cell proliferation. 1.25% , 2.5% and 5% medicated serum had significant effect on promoting endothelial cell migration or adhesion and inhibition the EphB4 expression. 1.25% and 2.5% medicated serum inhibited EphrinB2 expression. Conclusion XZD had significant effect on cell migration and adhesion to promote angiogencsis which is closely related to the EphB4/ EphrinB2.
