目的研究供体凋亡细胞预输注对大鼠胰岛移植后胰岛移植物功能的影响。方法应用STZ制备SD大鼠糖尿病模型,以直线加速器照射诱导供体Wistar大鼠睥细胞凋亡。经阴茎背静脉输注供体凋亡脾细胞,7d后于糖尿病SD大鼠双侧肾包囊进行同种异体胰岛移植,通过测定血糖变化、血清胰岛素水平以及胰岛移植物存活时间来观察胰岛移植物的功能状态。结果预输注供体凋亡脾细胞能显著延长同种异体胰岛移植物存活时间(中位生存时间达31d),并较长时间地维持受体鼠血清胰岛素处于较高水平。结论供体凋亡细胞预输注能够显著延长同种异体胰岛移植物的存活时间,能够较长时间维持胰岛移植物的正常功能状态。
Objective To observe the influence of transfusion of donor apoptotic cells on the function of islet grafts in rat islet transplantation. Methods Recipient SD rats were rendered diabetic by a single intraperitoneal injection of streptozotocin (STZ). Donor Wistar rat apoptotlc splenocytes were induced by X - irradiation with electron linear accelerator. The donor apoptotic splenocytes were injected into the recipient SD rats via dorsal veins of penis. Diabetic SD rats received islets transplantation under their renal capsule from the same donor rat 7 days later. The survival time and insulin level were examined to reflect the function of islet grafts. Results Pre - infusion of donor apoptotic splenocytes significandy prolonged allograft survival time ( MST 31 d). Serum insulin level in diabetic SD rats had a significant increase at 7d [ ( 19. 26 ± 4.50 ) mIU/ml ] and 14 d [ ( 20.70 ± 6.33 ) mIU/ml ] after transplantation in apoptotic group and decreased after rejection. Conclusion Transfusion of donor apoptotic splenocytes prolongs the survival and normal function of islet graft.
