目的 研究稳定表达Makorin环指蛋白1(MKRN1)基因对人宫颈癌细胞系Siha细胞的影响.方法 利用作者构建的稳定表达MKRN1基因的Siha细胞株,观察MKRN1基因对人端粒酶逆转录酶(hTERT)基因mRNA转录表达水平、端粒酶活性以及其对细胞周期的影响.结果 转染Siha细胞后,稳定表达MKRN1基因明显抑制细胞hTERT基因mRNA水平的表达,明显抑制细胞中端粒酶活性,并且G1期细胞明显增加,而S期细胞明显减少.结论 稳定表达MKRN1基因可特异性降低宫颈癌细胞中hTERT mRNA水平,抑制癌细胞内端粒酶活性,抑制宫颈癌细胞增殖,并且阻止宫颈癌细胞从G1期进入S期,促进细胞凋亡.

英文摘要:

Objective To stably transfect MKRN1 gene in Siha cell clones for studying the effect of MKRN1 on cells. Methods To use the experimental group constructed MKRN1 gene Siha stable expression cell lines,observed the MKRN1 gene transcription of hTERT gene mRNA expression level of telomerase activity and its effects on cell cycle. Results Transfected Siha cells, stable expression MKRN1 significantly inhibited the expression of hTERT gene mRNA levels,significantly inhibited telomerase activity, and G1 phase cells increased significantly, while the S-phase cells decreased significantly. Conclusion MKRN1 stable expression can specifically reduce hTERT mRNA level of cervical cancer cells, inhibit telomerase activity in cancer cells,inhibit cancer cell prolifer- ation,and prevent cancer cells from G1 phase lo enter S phase and from S phase to M phase,in particular,the former mainly to promute apoptosis.
