噬菌体展示(Phage display)是将外源蛋白质或多肽展示在噬菌体表面,而对应的编码基因整合在噬菌体基因组中的一项用来筛选特定蛋白质或多肽的技术。这一技术实现了基因型与表型的一一对应,使得特定外源蛋白或多肽及其编码基因的筛选简便易行。噬菌体展示技术作为一种新的淘选工具,在鉴定特异性蛋白、多肽,特别是新型细胞膜穿透肽应用中显现出广阔的应用前景,为鉴定新型靶向递药载体、实现药物的靶向投递奠定了基础。该文概述了噬菌体表达载体的分类以及淘选过程,利用噬菌体展示技术淘选新型细胞膜穿透肽的应用及其研究进展。
Phage display technology was a powerful tool for identification of cell targeting peptide. Protein or peptides variants are expressed on the outside of phage virion,while the genetic material encoding each variant protein( peptide) sequences were inserted in the phage genome. As a new kind of bio- panning tool,phage display technology was a novel candidate to identify homing cell penetrating peptide for targeted drug delivery. In this review,the phage vector classification and selection process were summarized. And the methods for identification of homing cell penetrating peptides by phage display technology and the prospect for further application were mainly introduced.
