目的探讨右归丸含药血清对骨髓间充质干细胞成脂分化相关基因表达及其甲基化影响。方法制备右归丸含药血清备用。无菌条件下,取SD大鼠骨髓间充质干细胞,贴壁法培养,传代。第二代贴壁细胞分组:(1)对照组:用含10%盐水的DMEM培养;(2)成脂诱导组:用成脂诱导条件培养基培养;(3)右归丸组:10%右归丸含药血清加成脂诱导条件培养基;分别干预3 d后用RT-PCR及甲基化特异性PCR法检测PPARγ和C/EBPα基因表达和甲基化状态。结果右归丸组PPARγ和C/EBPα的表达明显低于其他各组(P<0.05)。右归丸组PPARγ和C/EBPα甲基化上调,组间比较差异显著(P<0.05)。结论右归丸含药血清可通过去甲基化的表观遗传调控机制抑制骨髓间充质干细胞向脂肪细胞分化。

Objective To observe the effect of serum contained Yougui pill for adipogenic marker and methylation .Methods Un-der aseptic condition ,bone marrow mesenchymal stem cells were isolated from the femur and tibia of SD rats .The second generation of adher-ent cells were divided into normal control ,bone induction,Yougui pill groups.PPARγand C/EBPαgene expressions and methylation status were measured by RT-PCR and methylation specific PCR assay after 1 days.Results PPARγand C/EBPαmRNA expressions of Yougui pill group were obviously lower than those of other groups (P<0.05).The methylations of PPARγand C/EBPαwere up-regulated(P<0.05). Conclusions The serum containing Yougui pills could inhibit adipogensis differentiating of bone marrow mesenchymal stem cells by demeth -ylation mechanism .
