背景:骨折愈合不同阶段牵涉一大群差异表达基因。目的:研究骨折三期中早期“证”的本质及辨证治疗的分子生物学机制。方法:基于骨折早期模型,运用基因表达谱芯片技术分析骨折早期基因表达谱,并分析基于三期辨证治则的中药干预后基因表达谱的变化,筛选出活血化瘀治法的关键应答基因。结果:治疗骨折早期有显著差异表达的基因50个,其中上调基因22个,下调基因28个。结论:活血化瘀药可以促进细胞增殖,调节机体免疫功能,促进成骨细胞分化和细胞外基质形成,从而促进骨折愈合。
Background : Fracture healing in the different stages involves in a large number of differentially expressed genes. Objective : To investigate the nature of the syndrome in the early stage of fracture healing and molecular mechanisms of the treatment based on syndrome differentiation of TCM. Methods : Based on the early fracture model, microarray analysis of fracture early gene expression profiles was used. The changes in gene expression profiles were analyzed based on the three dialectical rules after the Chinese intervention, screening the critical response genes for method of activating blood circulation to dissipate blood stasis. Results : For the treatment of fractures in early stage, there are significant differences in the expression of 50 genes, including 22 up - regulated genes and 28 down - regulated genes. Conclusion : Chinese medicine of promoting blood circulation and removing blood stasis can promote cell proliferation and modulation of immune function ; promote osteoblast differentiation and extracellular matrix formation, so as to promote fracture healing.
