目的:比较绞股蓝(Gynostemma pentaphyllum)的乙醇提取物经微生物转化前后成分的变化。方法:采用德氏乳杆菌保加利亚亚种(Lactobacillus delbrueckii subsp. bulgaricus)作为实验的主要菌种,在43℃条件下,液体发酵6d,对转化前后绞股蓝乙醇提取物进行非小细胞肺癌A549细胞的抑制活性检测,利用高效液相色谱方法,对转化前后成分的变化进行分析,通过LCMS-IT-TOF鉴定转化前的2个主要成分及转化后的4个主要成分。结果:保加利亚乳杆菌对绞股蓝提取物的微生物转化产物比原绞股蓝提取物具有更强的抑制非小细胞肺癌A549细胞增殖作用。实验显示保加利亚乳杆菌能够使绞股蓝中的成分发生转化,主要转化物被鉴定为Gypenoside L、Gypenoside LI、达木林A及达木林B,可为天然产物中有效成分的转化提供一个新的研究思路。
Objective: To compare the components of the ethanol extract from Gynostemma pentaphyllum beforeand after microbial transformation. Methods: The ethanol extract of G. pentaphyllum was fermented in liquid for6 d using Bulgarian lactobacillus at 43 ℃. The anticytotoxic activity before and after microbial transformationwere tested using non-small cell lung cancer A549 cells. The chemical composition was identified by LCMS-ITTOF.Results: The ethanol extract of G. pentaphyllum transformed by Lactobacillus delbrueckii subsp. bulgaricusshowed more potent cytotoxic activity against A549 cells than that of the original extract. Some components weretransformed by microbial transformation and the major transformed components were identified as gypenosideL, gypenoside LI, damulin A and damulin B. Conclusion: More bioactive components of natural products may beobtained by microbial transformation.
