目的:观察大鼠实验性自身免疫性脑脊髓炎(EAE)模型脑、脊髓组织干扰素γ(IFN-γ)与基质金属蛋白酶-9(MMP-9)免疫组化的表达,探讨滋阴补肾法(左归丸)与温阳补肾法(右归丸)治疗EAE的作用机制。方法:用髓鞘碱性蛋白(MBP)免疫Lewis大鼠诱导EAE模型,120只大鼠随机分为正常组、模型组、激素组、左归丸组及右归丸组,每组24只。正常对照组和模型组给予生理盐水灌胃,3mL/(只·d);激素组免疫后给予生理盐水灌胃,3mL/(只·d),发病后改为醋酸泼尼松混悬液灌胃,5mg·kg-1·d-1;左归丸组免疫后给予左归丸混悬液,2mg·kg-1·d-1;右归丸组免疫后给予右归丸混悬液,3mg·kg-1·d-1。直至处死。分别于急性期(免疫后15d)和缓解期(免疫后27d)随机选取各组大鼠,取脑组织切片进行IFN-γ、MMP-9免疫组化染色。结果:在造模后15d和27d,模型组大鼠脑和脊髓IFN-γ、MMP-9表达染色IOD值均较正常组明显升高,差异有统计学意义(P〈0.05);各治疗组IFN-γ的IOD值在给药后减低,同模型组相比差异均有统计学意义(P〈0.05)。造模后左归丸组MMP-9表达较模型组明显减少,差异有统计学意义(P〈0.01)。造模后27d,左归丸组脑和脊髓组织MMP-9表达较模型组显著降低(P〈0.01),且左归丸组脑组织中MMP-9表达染色IOD值较免疫后15d显著降低,差异有统计学意义(P〈0.05);右归丸组MMP-9表达较免疫后15d明显降低,差异有统计学意义(P〈0.05)。结论:左归丸和右归丸可下调EAE大鼠中枢神经系统IFN-γ、MMP-9表达,其中左归丸可下调急性期MMP-9表达,右归丸可下调缓解期MMP-9表达来治疗EAE。
Objective: To observe the immunohistochemical expression of interferon- 7 (IFN- ~/), matrix metalloproteinase -9(MMP-9) in brain and spinal cord tissue, and explore the action mechanism of Zuogui Pills and Yougui Pills on rats with experimental autoimmune encephalomyelitis(EAE). Methods: Lewis rats were immunized with the myelin basic protein (MBP).120 Rats were grouped randomly into nomal group, EAE group, prednisone group, Zuogui Pills group and Yougui Pills group after post immunization (PI). Rats in normal group and EAE group were administered sodium, each 3ml/d, rats in prednisone-group were administered suspension of prednisone after developed clinical signs, each 5mg/kg, rats in Zuogui Pills group were administered suspension of Zuogui Pills, each 2g/kg, and rats in Yougui Pills group were administered suspension of Yougui Pills each 3g/kg. On days 15 and 27 PI, rats were killed and the immunohistochemical staining was performed on the sections of brain and spinal cord. Results: On days 15 and 27 PI, the expression of IFN-γ and MMP-9 in central nervous system of EAE group were higher than those in nomal group (P〈0.05). The expression of IFN-γ in brain and spinal cord tissue of prednisone group, Zuogui Pills group and Yougui Pills group was lower than that in EAE group (P〈0.05). On day 15 PI, the expression of MMP-9 of prednisone group and Zuogui Pills group were respectively lower than that in EAE group (P〈0.01). On day 27 PI, the expression of MMP-9 in brain and spinal cord tissue of Zuogui Pills group were significantly lower than that in EAE group (P〈0.01). In Zuogui Pills group, the expression of MMP-9 in brain tissue on day 21 PI was significantly lower than that on day 15 PI(P〈0.05). In Yougui Pills group, the expression of MMP-9 in brain and spinal cord tissue on on day 21 PI was lower than that on day 15 PI (P〈0.05).
