目的观察二黄方对实验性自身免疫性脑脊髓炎(EAE)模型大鼠外周血NK细胞和细胞因子的影响。方法70只Lewis大鼠随机分为4组:正常组、模型组、西药组和中药组。模型组、中药组和西药组用完全福氏佐剂(CFA)与豚鼠脊髓匀浆(GPSCH)制成的混合乳剂注射Lewis大鼠两后足掌皮下建立EAE模型;正常组和模型组灌服生理盐水;中药组灌服二黄方;西药组灌服醋酸泼尼松。每天记录各组临床症状评分,造模后第14天和第28天分别采集外周血,用液体芯片技术测定细胞因子;用流式细胞仪检测NK细胞。结果大鼠造模后第20天和第28天中药组和西药组临床评分平均值比模型组显著降低(P〈0.05);造模后第28天中药组和西药组外周血NK细胞比模型组均显著降低(P〈0.05);造模后第28天中药组外周血白细胞介素-15(IL-6)比模型组显著降低(P〈0.05);西药组造模后第14天外周血IL-18比模型组升高(P〈0.05);第28天干扰素-γ(IFN-γ)比模型组降低(P〈0.05)。结论二黄方和激素均可明显改善EAE大鼠临床症状,二黄方可能通过调节NK细胞、IL-6发挥治疗作用。激素可能通过调节NK细胞、IL-18和IFN-γ来治疗EAE。
Objective To observe the effects of Erhuang Formula on NK cells and cytokines in peripheral blood of experimental autoimmune encephalomyelitis (EAE) rats. Methods Seventy Lewis rats were randomly divided into 4 groups, including normal group ( n = 10 ), model group ( n = 20 ), western medicine group (n = 20) and Chinese medicine group (n = 20). All of the rats except those in the normal group were injected with the antigen consisted of guinea pig spinal cord homogenate (GPSCH) and complete Freund's adjuvant (CFA) in both postpedes to induce EAE. The normal group and the model group were administrated with normal sodium; the western medicine group with Prednisone; the Chinese medicine group with Erhuang Formula. Clinical score was recorded every day after inducing. On the day 14 and 28, the peripheral blood was collected. Cytokines( IFN-γ, IL-6, IL-18) were detected by Liquidchip; NK ceils were detected by flow cytometer. Results Clinical score of the Chinese medicine group reduced on the day 20 and 28; the western medicine group reduced from day 20 to day 28, all compared with the model group. On the day 28, NK cells reduced in peripheral blood of the Chinese medicine group and the western medicine group compared with the model group; there were no significant differences between the two treatment groups. In peripheral blood of rats in the Chinese medicine group, IL-6 reduced on the day 28 ; in the western medicine group, IL-18 increased on the day 14 and IFN-γ decreased on the day 28 ; all compared with the model group. Conclusions Both Erhuang Formula and Prednisone can ameliorate obviously the clinical symptoms in EAE rats. Erhuang Formula probably regulates immune system by reducing NK cells and IL-6 to treat EAE. Prednisone likely regulates NK cells, IL-18 and IFN-γ to treat EAE.
