目的 获得高效、稳定表达含有HIV-1主要结构蛋白gag、pol和env的病毒样颗粒的293细胞系。方法 选择合适的真核细胞表达质粒与带有抗性基因的载体,共转染293真核细胞,用含有抗性的培养基筛选出稳定表达细胞株。结果 HIV-1的gag、pol和env蛋白均在293细胞中获得了表达,且不同代次的293细胞中蛋白表达情况无明显差异。结论 成功筛选了稳定表达HIV-1病毒样颗粒的293细胞系,为进一步开发艾滋病疫苗奠定了基础。
Objective To screen the 293 cell line for stable and high expression of virus-like particles (VLPs) containing structural proteins gag,pol and env of HIV-1. Methods Co-transfect 293 ceil line with eukaryotic expression vector D-GPEi for HIV-1 gag,pol and env genes and plasmid pcDNA3. 1 carrying neomycin resistant gene,and screen the ceil lines for stable expression with G418 resistant medium. Results The major structural proteins gag,pol and env of HIV-1 were expressed in 293 ceils. The expression levels in screened eel lines of various passages showed no significant difference. Conclusion The 293 ceil lines for stable expression of VLPs of HIV-1 were successfully screened,which laid a foundation of development of HIV vaccine.
