活性炭在体胚发生过程中能够促进细胞的生长及发育。为进一步提高杂交鹅掌楸体胚发生效率,以两个基因型的杂交鹅掌楸愈伤组织为材料,在培养过程中添加磷酸碳、针剂碳和Sigma公司活性炭,通过统计体胚总数及子叶胚数量,测定培养基的pH和渗透压,分析活性炭的吸附性能等,以筛选适合杂交鹅掌楸体胚发生的活性炭类型,并探索不同活性炭影响杂交鹅掌楸体胚发生效率差异的原因。研究结果表明:针剂碳、磷酸碳处理下诱导获得的杂交鹅掌楸体胚总数和子叶胚数显著高于Sigma公司活性炭,且针剂碳的效果最佳。针剂碳的吸附性能适中,添加针剂碳的培养基高压灭菌后其pH和渗透压也适中,说明活性炭的吸附性能会影响培养基的pH和渗透压,从而影响杂交鹅掌楸的体胚发生。
Activated carbon is often used to improve cell growth and development in somatic embryogenesis. In this stud- y, three types of activated carbon, including phosphoric acid activated carbon, activated carbon for refinement of injec- tion and activated carbon purchased from Sigma were added respectively to the mediums for improving somatic embryo- genesis frequency of two genotypes of cell lines in Liriodendron hybrids. The number of total somatic embryos and cotyle- don embryos were counted, pH and osmotic pressure of the mediums were detected, and the adsorption ability of activa- ted carbon were analyzed to select the most suitable activated carbonl and to find their potential effects on somatic embry- ogenesis of Liriodendron hybrids. The results showed that the first two types of activated carbon induced more total somat- ic embryos and cotyledon embryos than the third type, especially the activated carbon for refinement of injection. The ad- sorption ability of activated carbon for refinement of injection was moderate among the three, and the pH and osmotic pressure of the mediums containing it were also temperate compared with the other two types of activated carbon. It was concluded from the results that the adsorption ability of activated carbon may change the pH and osmotic pressure of me- diums directly to influence the somatic embryogenesis of Liriodendron hybrids.
