目的评价IκBα突变型基因转染对永生化神经前体细胞株(INPCs)增殖、分化及部分炎性细胞因子分泌的影响。方法在已经建立的转染pcDNA 3.1及转染pcDNA3.1/IκBαM的INPCs的基础上,用MTT法绘制两种细胞株的生长曲线,用流式细胞仪检测其增殖指数,以5%小牛血清诱导分化后,采用Western blot法测定两种细胞株的分化情况,同时用realtime RT-PCR测定肿瘤坏死因子-α(TNF-α)mRNA、白细胞介素-1β(IL-1β)mRNA和白细胞介素-6(IL-6)mRNA的表达。结果与转染pcDNA 3.1 INPCs比较,转染pcDNA 3.1/IκBαM INPCs的增殖指数、IL-1βmRNA和IL-6 mRNA表达降低(P〈0.05),TNF-αmRNA表达差异无统计学意义(P〉0.05)。经血清诱导分化后,两种细胞株均大量表达胶原纤维酸性蛋白。结论IκBα突变型基因转染可减慢INPCs的增殖,并减少部分炎性细胞因子的表达,但对INPCs的分化无影响。
Objective To observe the effect of mutated IκBαgene transfection on the proliferation, differentiation and cytokine secretion of immortalized neural progenitor cell strain(INPCs).Methods After the establishment of mutated IκBαgene transfected INPCs and blank plasmid transfected INPCs,MTF staining and flow cytometry were used to draw the growth curve and measure the cell proliferation index.Western blot was carried out to observe the differentiation of the two cell strains after 5 % blood serum induced differentiation.The expression of TNF-αmRNA,IL-1βmRNA and IL-6 mRNA was measured by reahime RT-PCR.Results It was established by growth curve and cell proliferation index that the proliferation was slowed down in mutated IκBαgene transfected INPCs compared with blank plasmid transfected INPCs.After blood serum induced differentiation,glial fibrillary acidic protein was highly expressed in the two cell strains.There was no difference in the expression of TNF-αmRNA between mutated IκBαgene transfected INPCs and blank plasmid transfected INPCs.The expression of IL-1βmRNA and IL-6 mRNA was reduced in mutated IκBαgene transfected INPCs.Conclusion Mutated IκBαgene can slow down the proliferation of INPCs,reduce the expression of some cytokines,but don't influence the differentiation of INPCs.
