目前,手性材料的研究日渐升温,尤其在医药领域,众多单一对映体形式的手性药物不断被批准上市。氯霉素(CAP)作为广谱抗生素,临床应用广泛,现采用化学合成法可进行大量生产。制备CAP的前体2-氨基-1-(4-硝基苯基)-1,3-丙二醇(氯霉胺,ANP)具有特异的立体化学结构,可进行各种途径的结构修饰和优化,且由于其来源丰富、价格低廉,已成为手性化合物领域研究的热点。近年来,相关学者充分利用光学活性ANP多官能团的结构、理想的区域选择性及广泛的生物活性等特点,对其作了不同途径的衍生化,获得了大量衍生物,并将它们用作工业手性拆分剂、不对称反应的手性催化剂和助剂、各类基础化学反应底物、药物发现的活性先导化合物等,预示了ANP及其衍生物广阔的应用前景。该文对此类手性材料相关的化学生物学性质作一综述。
Recently,much more attention has been paid to the chiral materials,especially in the pharmaceutical field,where a number of the drugs with optical activities have been put into market increasingly.Chloramphenicol(CAP) is a widely used broad-spectrum antibiotic,which can be produced in quantity by chemical synthesis industrially.As the precursor of CAP,2-amino-1-(4-nitrophenyl) propane-1,3-diols(chloramphenicol bases) are stereochemically pure agents,which can be modified and optimized through numerous ways.Moreover,ANP have been studied intensely owing to its ready availability and remarkable economy.During these years,the relative researchers have devoted a lot to its optimization and got a variety of its derivatives which has been used in different fields,such as chiral resolution agents,chiral catalysts and auxiliaries for stereoselective synthesis,the reaction materials,lead compounds for discovering new drugs and so on recently.And all the applications have proved to be available,which predicts the light future for the further development of ANP and its derivatives.This review summarizes the characteristics of the biochemistry concerned.
