用同源克隆的方法和3′RACE技术,从日本晚樱(Prunus lannesiana)中克隆得到了1个AP1同源基因ClAP1的cDNA全长。其cDNA全长1005bp,包括1个编码238个氨基酸共717bp的开放阅读框。序列比对和分子系统发生分析表明,ClAP1是拟南芥的AP1同源基因,其蛋白质的C末端具有一个保守的euAP1模体。半定量RT-PCR分析表明,ClAP1在3个日本晚樱品种‘大岛’、‘一叶’和‘普贤像’的萼片、花瓣、雄蕊和雌蕊中均有表达,在叶中不表达,属参与花发育的转录因子,但其表达模式与拟南芥的AP1有一定差别。
MADS-box genes encode transcriptional regulators that are critical for flowering and floral organogenesis. To find genes involved in the floral organogenesis of cherry blossom(Prunus lannesiana), cDNAs for one MADS- box genes,ClAP1,was cloned using homologous cloning and 3′ -RACE method based on the sequence database of NCBI. The full length of ClAP1 cDNA is 1 005 bp,containing an open reading frame(ORF)of 717 bp and coding for a polypeptide of 238 amino acid residues. Subsequently, sequence comparison and phylogenetic analysis revealed that ClAP1 was highly homologous to protein AP1 in Arabidopsis and belonged to the euAP1 clade of class A. However,semiquantitative RT-PCR analysis showed that ClAP1 was expressed in sepal, petal, stamen and carpel of three cherry blossom cultivar‘sMakino’,‘Hisakura’and‘Albo-rosea’,which was slightly different from the expression pattern of AP1.
