目的探讨结缔组织生长因子对人增生性瘢痕成纤维细胞磷脂酰肌醇3激酶/蛋白激酶B的激活作用。方法应用免疫印迹技术检测10ng/mL结缔组织生长因子作用于人增生性瘢痕成纤维细胞不同时间(0、5、15、30、60、120min)后蛋白激酶B的活化情况(以磷酸化蛋白激酶B和总蛋白激酶B比值表示),以及用同法检测磷脂酰肌醇3激酶抑制剂LY294002对结缔组织生长因子活化蛋白激酶B的作用。结果10ng/mL结缔组织生长因子作用于人增生性瘢痕成纤维细胞15min时,蛋白激酶B的活化水平(0.614±0.015)已明显高于对照组(0.308±0.021,P〈0.05),30min时最明显(0.724±0.017,P〈0.01),而且持续至少达120min,与对照组相比较,差异有统计学意义。10μmol/L LY294002作用人增生性瘢痕成纤维细胞后蛋白激酶B的活化水平下降(0.135±0.025),同对照组(0.422±0.015)相比较,差异有统计学意义(P〈0.05)。结论结缔组织生长因子可激活人增生性瘢痕成纤维细胞磷脂酰肌醇3激酶/蛋白激酶B信号通路,LY294002可抑制结缔组织生长因子对此通路的激活。
Objective To explore the activation of phosphatidylinositol 3-kinase/ protein kinase B(PI3K/PKB) induced by connective tissue growth factor in human hypertrophic scar fibroblasts. Methods Levels of phosphorylated PKB and total PKB were examined by western blotting in human hypertrophic scar fibroblasts after stimulation with connective tissue growth factor at 10 ng/mL for different time periods(0,5,15,30,60,120rain). Inhibitory effect of LY294002 on activation of PKB induced by connective tissue growth factor was also examined, p-PKB/ PKB ratio was used to indicate levels of activation of PKB, Results p-PKB/ PKB at 15min (0.614 ±0.015) was significantly higher than the control group (0. 308±0. 021 ,P〈0. 05), Maximal activation of PKB (maximal levels of p-PKB/PKB) took place at 30min (0. 724±0. 017 ,P〈0. 01) and the activation persisted at least 2h, PI3K inhibitor LY294002 could inhibit the activation of PKB induced by connective tissue growth factor in human hypertrophic scar fibroblasts, Levels of PKB (p-PKB/ PKB) decreased (0.135±0.025) with concentrations 10μmol/ L of LY294002 (P〈0, 05) ,compared with the control group(0.422±0,015), Conclusion Connective tissue growth factor can activate PI3K/PKB signaling pathway in human hypertrophic scar fibroblasts, This action of connective tissue growth factor is PI3K dependent and can be inhibited by LY294002.
